There's been a lot of questions about the karma seal so today I'm gonna be dropping some theories that might answer your questions for you.

 First of all let's talk about how Boruto got his jougan.

Boruto jougan

Boruto jougan is a type of dojutsu which haven't been seen before in the series but the eye was recognizable by the otsutsuki (urashiki and momoshiki). It has been noticed that Boruto can't activate it at will, it only activated when Boruto is in a situation of intense danger, so for now we can say Boruto doesn't have control over the eye .

Why does Boruto have the jougan

Why Boruto possesses the jougan is yet to be found out but this question leads to another question, why did Boruto awaken the jougan ?.

Now you all can remember momoshiki saying you have inherited otsutsuki power in their  final fight   So why did Boruto inherit otsutsuki power? Well the answer is his blood line.

Now come to think of it , otsutsuki powers was past down to some clans by generations. For example, the uchiha clan inherited Indra's sharingan and the hyuga clan inherited otsutsuki byakugan . Now Hinata (Boruto's mom) has the byakugan and she's the the byakugan princess and also during Boruto the last, Hamura gave her some fraction of his chakra, so I'm conclusion, Hinata has strongly inherited otsutsuki power. Now coming to Naruto (Boruto's dad) his. Recarnation of Ashura which is Hagoromo's son and Hagoromo gave Naruto half of his chakra during the 4th great ninja war making him to activate six paths sage mode and his also a partial jinchuriki of the ten tails so he has inherited  otsutsuki chakra to. In conclusion both Boruto parents has otsutsuki chakra in them . So how did Boruto awaken the jougan ? Well to me Hagoromo chakra combines with Hamura chakra makes owner to awaken the jougan , since Naruto has Hagoromo chakra and Hinata has Hamura chakra so definitely by birth, Boruto has both chakra . Just like how Indra chakra combines with Ashura chakra makes the owner to activate the reningan.

So this case is settled. Let's move further.

Well there's a truth behind why Boruto and kawaki has this curse seal. Let's take a good look at 

Boruto curse seal.

Boruto curse seal started with his hand and it started spreading with some strange markings . The karma seal is said to be able to store 100 ninja jutsu as well as be able to use them that means that Boruto can store and use 100 ninja via the karma seal. Recently in the manga it was seen that Boruto's karma seal can absorb jutsus and he used it to absorb a fire jutsu placed on konohamaru. Remember the karma seal can absorb and store jutsu that means Boruto can use the stored jutsu which is similar to momoshiki reningan, one hand absorbs the other releases it. So could the curse seal be working with the jougan ? Well I take yes for an answer because the jougan and the karma seal originated from the otsutsuki so that means the jougan is a higher version of  reninsharingan just like how the reningan is a higher version of the Mangekou sharingan .

Why does Boruto have the karma seal?

The karma seal appeared on Boruto when he defeated momoshiki otsutsuki, and momoshiki appeared to him and said:  

"You have inherited an otsutsuki power". 

What does this statement means?  Does it means he inherited it because his parents are descendants of the sons of kaguya ( Hagoromo and Hamura Otsutsuki)? Or is it because he defeated an otsutsuki? Or is it because momoshiki gave it to him because he defeated him ? Well we don't know for now but it would be seen later in the series.

Does Boruto jougan work with his karma seal?

 As you can see the curse seal mark which started from his hand has spreads to his face . Now see what happens when he uses the karma seal at this point 

 As you can see, the curse mark seal spreads to his eye  and give him a marking So maybe, just maybe Boruto jougan and his karma seal works together .

Now let's take a look at kawaki curse seal.

 Kawaki karma seal

As you can see , kawaki also has the mark which runs from this hand to his face just like Boruto. But in this case, his markings turns red mean while Boruto markings turns blue. This means something 

but it hasn't been revealed yet

 Boruto and kawaki karma seal both has similar powers but I think there is a slight difference between them. 

 For now we don't know what his intentions are but we will soon find out later in the series.Within time we would come to notice it as the series goes on.
